Biosynthesis and crystallization mechanism of chitin microfibrils

Chitin is considered the second most abundant biopolymers on earth next to cellulose. While the majority of cellulose is distributed in the plant kingdom, chitin can be found in various algae, fungi, invertebrates, and even some vertebrates. They utilize chitin mainly as structural components for its outstanding mechanical properties (i.e. fungal cell wall, crustacean exoskeleton). Chitin is biosynthesized in a fibrous crystalline form, namely microfibril, having a diameter of a few to tens of nanometers, and a length of typically a micrometric order. The microfibrillar morphology varies depending on its origin. Although the crystal structures of chitin allomorphs have been investigated by diffraction techniques, their ultrastructural aspects as well as structure-properties relationship have been poorly documented until recently, compared to other cellulosic crystals.
In this talk, I will share our recent studies on ultrastructure and structure-properties relationships of chitin allomorphs using a combination of experimental and computational techniques. The ultrastructural details will be discussed in association with its biosynthetic mechanisms.
